The human immune system can directly lyse invading micro‐organisms and aberrant host cells by generating pores in the cell envelope, called membrane attack complexes (MACs). Recent studies using single‐particle cryoelectron microscopy have revealed that the MAC is an asymmetric, flexible pore and have provided a structural basis on how the MAC ruptures single lipid membranes. Despite these insights, it remains unclear how the MAC ruptures the composite cell envelope of Gram‐negative bacteria. Recent functional studies on Gram‐negative bacteria elucidate that local (...) assembly of MAC pores by surface‐bound C5 convertase enzymes is essential to stably insert these pores into the bacterial outer membrane (OM). These convertase‐generated MAC pores can subsequently efficiently damage the bacterial inner membrane (IM), which is essential for bacterial killing. This review summarizes these recent insights of MAC assembly and discusses how MAC pores kill Gram‐negative bacteria. Furthermore, this review elaborates on how MAC‐dependent OM damage could lead to IM destabilization, which is currently not well understood. A better understanding on how MAC pores kill bacteria could facilitate the future development of novel strategies to treat infections with Gram‐negative bacteria. (shrink)

Die drei Bücher "Vom rechten Handeln" beschäftigen sich mit dem Verhältnis zwischem dem "Sittlichen" und dem "Nützlichen". Cicero vertritt die These, dass beide Begriffe "von Natur aus" identisch sind. Einen Konflikt zwischen dem Sittlichen, d.h. den aus den Tugenden folgenden Pflichten, und dem Nützlichen kann es daher im Grunde nicht geben. Was immer auf den ersten Blick sich als Konfliktfall darstellen mag - und Cicero geht zahlreiche Beispiele durch -, erweist sich bei genauer Betrachtung stets als scheinbarer Konflikt, der Nutzen (...) als scheinbarer Nutzen. Cicero verfasste "De officiis" als Vermächtnis an seinen Sohn in den Monaten nach der Ermordung Caesars. Von allen Werken Ciceros hat es am stärksten auf die Nachwelt gewirkt, von der Antika an, über Mittelalter, Renaissance bis hin zur Neuzeit; es wurde 1465 in Mainz als erstes gedruckt und am häufigsten übersetzt und kommentiert. Voltaire und Friedrich der Große hielten "De officiis" für das beste Buch über Moral. Die Übersetzung Karl Büchners sucht so wörtlich wie möglich die kunstvolle Sprache Ciceros nachzubilden. Der lateinische Text folgt der maßgeblichen Ausgabe von C. Atzert, die zahlreichen Abweichungen werden ausführlich kommentiert. Mit Einführung, Verzeichnis der Eigennamen und Literaturhinweisen. Methoden. Im obigen Artikel ist die „Phylogenie des Stammbaumes” untersucht worden. Beginnend mitHaeckel werden 26 Typen phylogenetischer Symbole kritisch besprochen, d.h. nicht die Resultate, sondern nur die Methoden, z.B. bezüglich Systematik und Phylogenie, lebender und ausgestorbener Organismen, geologischer Perioden, Stufen und homologer Variationen, geographischer Verbreitung, Diversität, etwaiger Bedeutung der Einzelheiten, Mono-, Bi- und Polyrheithrie , usw. Der Faktor Zeit wird dabei für phylogenetische Systeme als der wesentlichste betrachtet. Der Autor hat daher in seinen- neuen Darstellungen die Begriffe „Zeit-Stufen” oder „Zeit-Globen-Oberflächen” (...) eingeführt; diese stehen senkrecht zum „Zeit-Radius”. Auf diesem Grunde wird ein neues Kriterium zur Unterscheidung systematischer und phylogenetischer Vorstellungen denjenigen anderer Autoren hinzugefügt. Erläuterungen. Nach einer Übersicht über die besprochenen Schemen im 1. Teil werden die Verhältnisse zwischen systematischen und phylogenetischen Darstellungen sowie die Verbindung ihrer Symbole untersucht. Es wird gezeigt, dass viele Einzelheiten solcher Konstruktionen oft, wissentlich oder unwissentlich, keine bestimmte Bedeutung haben. Der Verfasser hat versucht zu zeigen, dass es erwünscht wäre, möglichst mit allen Einzelheiten einer symbolischen Darstellung irgendeine Bedeutung zu verbinden: Linien und Entfernungen in der Ebene, Winkel in räumlichen Schemen. Die letztgenannten werden besonders für solche Arten von phylogenetischen Systemen empfohlen, bei denen der Faktor Zeit stets eine der Dimensionen bildet. Dieser Grundsatz wird mit der Evolution systematischer Einzelheiten in Verbindung gebracht, und Merkmals-Phyletik, homologe Variationen, Stufen, Polyrheithrie, Analogie und Homologie, Konvergenz und Verwandtschaft, etc. werden kurz im Lichte der Vererbungslehre besprochen. Es istHayata's extrem dynamischen Ansichten zu verdanken, dass das Statische in den systematischen und phylogenetischen Darstellungen aufgelockert worden ist. Méthodes. Dans l'article inséré ci-dessus il s'agit de ce qu'on pourrait appeler „la phylogénie de l'arbre généalogique”. En commençant parHaeckel , l'auteur discute 26 types divers de symboles phylogénétiques, c.à.d. seulement les méthodes , par exemple en ce qui concerne la taxonomie et la phylogénie, les organismes récents et éteints, les périodes géologiques, les niveaux et les variations homologues, la distribution géographique, la diversité, la signification éventuelle des détails, la mono-, bi- et polyrheithrie , etc. Le Temps est considéré comme l'élément le plus essentiel dans tout système phylogénétique. Par conséquence, l'auteur a introduit dans ses nouvelles représentations les idées de „Niveaux” ou de „Surfaces des Sphères du Temps” qui sont imaginés perpendiculaires au Radius-Temps. Dans l'ordre de ces idées un nouveau critère est ajouté à ceux donnés par d'autres auteurs pour la distinction des systèmes taxonomiques et des représentations phylogénétiques. Justifications. Après une récapitulation des systèmes, énumérés dans la première partie, les relations des constructions taxonomiques et phylogénétiques, ainsi que les connections de leurs symboles sont discutées. Souvent plusieurs détails sont, sciemment ou insciemment, dépourvus de toutes signification. L'auteur a essayé de démontrer qu'il serait désirable d'attribuer autant que possible de significations définies au détails d'une représentation symbolique de la nature décrite: aux lignes et aux distances dans le plan, aux angles dans l'espace. Les constructions tridimensionales sont particulièrement recommandées pour plusieurs systèmes phylogénétiques; dans de tels systèmes le Temps devrait toujours remplir la fonction d'une des dimensions. Ce principe est mis en rapport avec l'évolution des unités taxonomiques; phylogénie des charactères, variations homologues, niveaux, polyrheithrie, analogie et homologie, convergence et relations héréditaires, tous ces sujets sont brièvement discutés dans leurs rapports avec la Génétique. Selon l'opinion de l'auteur, c'est aux idées ultra-dynamiques d'Hayata que la taxonomie et la phylogénie doivent la pénétration féconde du dynamisme dans leurs systèmes trop souvent statiques. A theme like “town and science” invites to comparative analysis, and suggests questions like these: Is the urban context a particularly fertile soil for the development of scientific thinking? Or rather the contrary? Is it fertile or barren under specific circumstances? Or does it favour a particular kind of scientific activity?General answers to such questions can hardly be found; still, they may provide case studies with a guiding perspective. Case studies, on the other hand, may lead to better understanding of (...) the implications of the general questions if not to answers. In the following I therefore try to read the emergence of Mesopotamian mathematics into the context of the “urban revolution” and the rise of states (c. 3300 to c. 2300 B.C.).Mesopotamian city states appeared in the late fourth millennium B.C., and were eventually absorbed in larger territorial states a thousand years later. Contemporary with the incipient state formation was the creation of a script and of genuine, coherent mathematics. Both were based on earlier accounting techniques, both were used by Temple officials for administrative purposes, and both were marked decisively by this application. At the same time, both presuppose modes of thought which are not produced directly by their application but only as mediated by a school system.In the mid‐third millennium, an autonomous scribal profession appears for the first time. At the same time emerge the first literary texts and the first “pure” mathematical problems (i.e., problems not directed at or following the pattern of genuine application). It seems as if the scribes, as soon as they were emancipated as a craft, tried out the range of their professional tools.Neither the use of writing for literary purposes nor the possibility to use numbers for “higher” computation were forgotten when the city states were absorbed in larger social systems. Both, however, were subsumed by the interest of the new territorial state, the former as a means to propagate royal ideology, the latter in the royal administration.The final section of the article compares this early Mesopotamian development with other instances of the “urban revolution” and with the development of scientific thought in the Ancient Greek city state and in the context of Medieval city communes. The fundamental tenet of contemporary sociobiology, namely the assumption of a single process of evolution involving the selection of genes, is critically examined. An alternative multiple-level, multiple-process model of evolution is presented which posits that the primary process that operates via selection upon the genes cannot account for certain kinds of biological phenomena, especially complex, learned, social behaviours. The primary process has evolved subsidiary evolutionary levels and processes that act to bridge the gap between genes and these complex behaviours. The (...) subsidiary levels are development, individual animal learning, and socioculture itself. It is argued that individual learning is pivotal to the derivation and biological analysis of culture. The differences between cultural and noncultural societies are stressed. It is concluded that such a multiple-level model of evolution can form the basis for reconciling opposing sides in the sociobiology debate. (shrink)
